It was a flawless 18 holes from Clarke as he shot a seven under par 65.
Carding birdies on the second, sixth and ninth, Clarke shot a blistering three under par front nine.
And Lowry had an eagle on the par five 10th as well as birdies and 3 and 15.
Clarke is tied with Kevin Sutherland at the top of the leaderboard.
And hot on their heels are Corey Pavin, Mike Weir, Justin Leonard, Alex Cejka and Ernie Els.
