As of 10.30pm on Friday, the fire was still going on.
The Chiltern Firehouse is a luxury five-star hotel featuring 26 suites and an award-winning restaurant.
Located on Chiltern Street, it occupies a historic Victorian fire station, originally built in 1889 as the Chiltern Street Fire Station.
The hotel is a celebrity hotspot and has been visited by A-listers including Brad Pitt, Tom Cruise, Jennifer Lawrence, Lady Gaga and Johnny Depp.
The blaze broke out just hours before Valentine's Day dinner service.
A plant room on the roof level is also alight. The cause of the fire is not known at this stage, say the London Fire Brigade.

London Fire Brigade said: ‘Twenty fire engines and around 125 firefighters have been called to a fire at a hotel on Chiltern Street in Marylebone.
“Around one hundred people have evacuated the building before the Brigade arrived.
“The Brigade's 999 Control officers were first called at 1452 and deployed fire crews from Euston, Soho, Paddington, Soho, West Hampstead, Kensington, Chelsea and surrounding fire stations and are still at the scene.
“The cause of the fire is not known at this stage.”

Owned by hotelier André Balazs, who also owns the famous Chateau Marmont in LA, the Chiltern Firehouse is an exclusive social hub. There is also a private entrance for VIPs.
Mr Balazs said in a statement: “Earlier this evening, a fire broke out at Chiltern Firehouse. We are grateful to confirm that no one was hurt, and our guests and staff safely evacuated. The fire has now been fully contained.
“It is with heartfelt gratitude and appreciation that we watched a remarkable 140 fire fighters from over 20 stations rapidly descend on what they told me was a hugely sentimental building for so many of them.
“We know in fact one of those who rushed to the Chiltern Firehouse this evening had been stationed in the building when it was a fire station 30 years ago. I am truly grateful to all of them as I am sure that this is not the Valentine's Day evening they had in mind.
“As of now, as this is still a live situation, we do not have confirmation of what may have caused the fire, but the fire services are investigating.

“In the meantime, the Chiltern Firehouse will remain closed until further notice.”
